**Background**
Sphingosine-1-phosphate (S1P) receptors are a family of G protein-coupled receptors that play critical roles in regulating lymphocyte trafficking and immune responses. Among these, S1P1 is primarily responsible for the egress of lymphocytes from lymphoid organs into the blood and lymph. By modulating S1P1 activity, it is possible to sequester lymphocytes within lymph nodes, thereby reducing the infiltration of inflammatory cells into target tissues. This mechanism makes S1P1 an attractive target for treating autoimmune diseases and preventing organ transplant rejection. However, achieving selectivity over other S1P subtypes, particularly S1P3, is essential to avoid potential cardiovascular side effects. In this context, we will introduce a potent and S1P3-sparing S1P1 agonist – CS-2100.
**Definition**
CS-2100 is a potent, selective, and orally active S1P1 agonist with an EC50 value of 4.0 nM for the human S1P1 receptor.
**In Vitro and In Vivo Studies**
According to the CS-2100 description, this compound (also known as Compound 10b) is designed to provide high selectivity for S1P1 while sparing S1P3. In terms of CS-2100 biological activity, in vitro assays using CHO-K1 cells expressing the receptors demonstrated that CS-2100 acts as a potent agonist at both rat S1P1 (EC50 = 1.5 nM) and human S1P1 (EC50 = 4 nM), as assessed by the induction of GTPγS binding. Conversely, it showed significantly lower activity at the rat S1P3 receptor (EC50 = 7400 nM) and virtually no activity at the human S1P3 receptor (EC50 > 20,000 nM).
Furthermore, CS-2100 in vivo studies have validated its therapeutic potential. In rat models of heart valve graft rejection (HvGR), CS-2100 demonstrated significant immunosuppressive efficacy with an ID50 (infective dose) of 0.407 mg/kg. These results indicate that the compound can effectively modulate immune responses through oral administration. In conclusion, CS-2100 is a potent, orally active, and S1P3-sparing S1P1 agonist that holds promise for the development of selective immunosuppressive therapies.
